Morphology

• Acid fast (Z-N positive), rod shape, aerobic, non motile

and non-spore forming.

• Cytochemically : Gram +Ve

• Mycobacteria means “ Fungus bacterium” because some

time mycobacterium show branching filamentus

resembling to fungus mycelium in liquid medium.

• Bovine TB can be transmitted from livestock to

humans, deer and other animals

Diagnosis of Bovine TB

• Ziehl Neelsen staining

 Principle: Heat fixed impression smears are stained with

carbol fuchsin, rinsed and counterstained with malachite green.

Interpretation: Acid fast organisms (mycobacteria) retain

their red color and are distinguished from other material and

microorganisms which absorb the counter stain.

• M. bovis isolates – Stone brink’s medium

• M. tuberculosis & M.avium – Lowenstein Jensen slopes
