Mycobacteria
➢1882 Robert Koch discovered tubercle bacilli
➢Ehrlich discovered acid fast property of mycobacterium
➢John described johne’s bacilli
CLASSIFICATION OF
MYCOBACTERIA
Mycobacterial (acid-fast) cell wall
CLASSIFICATION OF
MYCOBACTERIA
3. Non Cultivable Mycobacteria
Mycobacteium leprae
Cell wall structure
Morphology
• Acid fast (Z-N positive), rod shape, aerobic, non motile
and non-spore forming.
• Cytochemically : Gram +Ve
• Mycobacteria means “ Fungus bacterium” because some
time mycobacterium show branching filamentus
resembling to fungus mycelium in liquid medium.
Biochemical reaction
Mycobacterium bovis
• Bovine TB can be transmitted from livestock to
humans, deer and other animals
Diagnosis of Bovine TB
• Ziehl Neelsen staining
Principle: Heat fixed impression smears are stained with
carbol fuchsin, rinsed and counterstained with malachite green.
Interpretation: Acid fast organisms (mycobacteria) retain
their red color and are distinguished from other material and
microorganisms which absorb the counter stain.
• M. bovis isolates – Stone brink’s medium
• M. tuberculosis & M.avium – Lowenstein Jensen slopes
3. Animal inoculation
•M. bovis- Guinea pigs, Rabbits
•M. tuberculosis- Guinea pigs, Rabbits
•M.avium- Chicken,Rabbit
Tuberculin test .
Immuno-diagnosis of tuberculosis
• Leprosy in human
• M.leprae grow on foot pad of mice and Nine
banded armadillo
PARATUBERCULOSIS
➢Johne and Frothingham (1894) first described
Paratuberculosis
➢Acid fast rods , found in clump (Bunch of faggot)
mycobactin killed extract of M. phlehi o
Sheep’s Prolapse of colon in Sheep
Enlarged mesentric lymph node
